MEDIPAL HOLDINGS Enters into Agreement with JCR Pharmaceuticals for JR-479, a Novel Therapy for GM2 Gangliosidosis

MEDIPAL HOLDINGS and JCR Pharmaceuticals have entered into an exclusive global licensing agreement and a co-development and commercialisation partnership in Japan for JR-479, an investigational treatment for GM2 gangliosidosis, an ultra-rare lysosomal storage disorder.
GM2 gangliosidosis results from a deficiency of the enzyme β-hexosaminidase A (Hex A). Depending on which subunit of Hex A is affected, the disease presents as Tay-Sachs or Sandhoff. The accumulation of GM2 gangliosides and related lipids leads to severe neurodegeneration. In the most severe cases, around half of affected children do not live beyond the age of three. With no approved treatments available, there is a critical need for new therapeutic options.
GM2 gangliosidoses are inherited conditions caused by genetic mutations that prevent the normal breakdown of GM2 gangliosides in lysosomes, resulting in harmful accumulation, particularly in nerve cells. Tay-Sachs disease and Sandhoff disease are the two most recognised forms. The disorder affects an estimated one in 300,000 people worldwide, is life-threatening, and currently has no approved therapies.
JR-479 has been developed using JCR’s proprietary J-Brain Cargo® platform, a technology designed to deliver medicines across the blood–brain barrier. This platform was first clinically validated with IZCARGO®, which was approved in Japan in 2021 for another lysosomal storage disorder. Preclinical studies have shown that JR-479 reaches the central nervous system, reduces disease-related substances, and improves survival in animal models.
Under the licensing agreement, MEDIPAL will hold exclusive rights outside Japan to develop, manufacture, and commercialise JR-479, including sub-licensing rights. Within Japan, MEDIPAL will support clinical trial logistics, awareness initiatives, and operational activities, while JCR will lead commercialisation and record domestic revenues.
