Repare Therapeutics Enters into Agreement with Debiopharm for Global Rights to Lunresertib

Repare Therapeutics has entered an exclusive global licensing agreement with Debiopharm for its investigational cancer therapy, lunresertib. 

Lunresertib is a first-in-class PKMYT1 inhibitor designed to target hard-to-treat solid tumours.

This licensing agreement builds on an earlier clinical collaboration between the two companies, which explored the combination of lunresertib and Debio 0123 a selective WEE1 inhibitor developed by Debiopharm. With this new arrangement, Debiopharm will assume full responsibility for the MYTHIC clinical study and will lead all current and future development activities involving lunresertib.

Under the terms of the agreement, Repare will receive an upfront payment of US$10 million. The company is also eligible to receive up to US$257 million through potential clinical, regulatory, commercial, and sales milestones, including up to US$5 million in near-term payments. Additionally, Repare will be entitled to single-digit royalties on global net sales.

The agreement follows promising early-phase data suggesting the combination of lunresertib and Debio 0123 may have a synergistic effect, offering potential therapeutic benefits for patients with cancers that are difficult to treat.

The POLAR trial is investigating RP-3467, a small molecule Polθ inhibitor. This compound is designed to exploit synthetic lethality in cancers associated with BRCA mutations and related genomic alterations. The ongoing trial is assessing the therapy alone or in combination with olaparib in adults with advanced ovarian, breast, prostate, or pancreatic cancers. Initial data on safety, tolerability, and early efficacy is expected in Q3 2025.

The LIONS trial is evaluating RP-1664, an oral, highly selective PLK4 inhibitor linked to TRIM37 overexpression. This first-in-human study is assessing its safety, pharmacokinetics, and early efficacy in patients with solid tumours. Early topline results from the trial are anticipated in Q4 2025.

Through this licensing deal, Repare aims to streamline its clinical priorities while ensuring the continued development of lunresertib through its partnership with Debiopharm.
