This poster demonstrates a scalable bioreactor-based platform for expanding cord blood-derived hematopoietic stem cells (HSCs) and differentiating them into NK cells for allogeneic immunotherapies. Using the Ambr® 15 system, optimized cytokines, and IBR403, the team achieved efficient CD34+ cell expansion and established a robust manufacturing approach for future cell therapy production.
Key Learning Points:
- Bioreactor-based HSC expansion offers a more scalable alternative to traditional 2D cell culture methods.
- IBR403 and DOE optimization improved CD34+ cell expansion while reducing cytokine requirements.
- Expanded HSCs can be differentiated into NK cells, supporting off-the-shelf therapies for cancer and immune disorders.
