MHC Class II Ultimers™ ProImmune manufactures MHC Class II Ultimers™, a multimer technology with a superior peptide-linked planar design and optimized fluorochrome for immunologists who wish to detect and quantitate CD4+ antigen-specific T cells.

ProImmune delivers custom Class II Ultimers™ quickly, in weeks rather than months, with stringent quality control and low batch-to-batch variability. It is a fully compatible technology; the staining procedure is substantially the same as that for other MHC tetramers. The staining protocols for the use of Class II Ultimers™ are essentially the same as those for Class II tetramers.

Consistent with our Pro5® MHC Pentamers, Class II Ultimers™ are available as custom made and off the shelf products. Similar to Class I Pentamers, custom Class II Ultimers™ are available in 6 weeks. ProImmune's Class II Ultimer™ technology is affordable and prices are comparable with the existing Pentamer technology.

Analogous with MHC Pentamers, MHC Class II Ultimers™ are aligned in a planar configuration, optimized for best access of the T Cell Receptors (TCRs) on the cell surface to the MHC binding groove. Class II Ultimers™ are linked to a core structure through their peptides, which means that each MHC molecule can independently align with its complementary TCR, improving the binding characteristics. In contrast, in a standard tetramer all MHC molecules point away from the central streptavidin core and therefore their peptide binding grooves do not face in the same direction.

The labeling technology for Class II Ultimers™ has been optimized in order to achieve the best combination of signal strength (brightness) and low background (non-specific) staining. Class II Ultimers™ are available labeled with R-PE and APC.
